What is the difference between NAD+ and NMN?
NAD+ is the active coenzyme that cells use to run energy metabolism. NMN is a precursor — a smaller molecule the body converts into NAD+ through the salvage pathway. Every NMN supplement is an indirect way of raising NAD+. Taking NAD+ itself skips a conversion step.

What is NMN?
NMN is nicotinamide mononucleotide, a nucleotide derived from ribose and nicotinamide. The body makes NMN from nicotinamide via the enzyme NAMPT, or from nicotinamide riboside (NR) via NR kinases. NMN is then converted into NAD+ by NMNAT enzymes. It is one step away from NAD+ in the salvage pathway.
NMN and NR are the two precursors that have received the most research attention. The 2018 Cell Metabolism review of NAD-boosting molecules summarises the in vivo evidence for both, and a 2024 review on NAD+ precursors gives a more recent picture of where the science stands.
Does NAD+ decline with age?
Yes. Measured NAD+ levels fall with age across multiple tissues in both animal models and humans. This decline is one of the main reasons NAD+ and its precursors became a research focus in the first place, and it is well documented in the reviews cited above.
What is not settled is the practical consequence for a healthy adult taking a supplement. Much of the most striking data comes from mice, and rodent findings do not transfer automatically to people. Treat any source that presents animal results as a human outcome with caution.
Is NMN or NAD+ better to take?
There is no consensus answer, and anyone claiming one is settled is overstating the evidence. The honest summary is that they work at different points in the same pathway: NMN relies on your body's conversion enzymes, while NAD+ supplies the coenzyme form directly. Both approaches are used in commercial formulas.
The argument for precursors is that smaller molecules may be absorbed more readily. Is NMN legal in the United States?
Yes, as of September 2025. NMN's regulatory status changed twice, which is why older articles contradict each other. In November 2022 the FDA determined NMN was excluded from the dietary supplement definition under the drug preclusion clause, because it had been authorised for investigation as a new drug first.
